The film opens with a couple engaging in sex. Afterwards, the man showers while the woman goes to get herself a drink. After his shower, he discovers "You're Next" written in blood on the sliding glass doors; behind them is the woman's dead body. He is then killed by an unknown assailant.
A wealthy family go to their remote vacation house. The mother, Aubrey (Barbara Crampton), claims she hears something upstairs and wants to leave. Ignoring her warning, Paul (Rob Moran) goes to investigate, but is startled by his son Crispian (A.J Bowen).
The next day, the family's other siblings arrive. Aubrey sends Erin (Sharni Vinson) over to the neighbors' house to ask for some items she needs, but there is obviously no response as they were murdered the night before. That night, at dinner, the family gathering quickly turns deadly when Tariq (Ti West) is shot with a bolt through a window. In the process of escaping the room, Drake (Joe Swanberg) is shot in the back with a bolt and passes out from the pain. The daughter of the group, Aimee (Amy Seimetz), attempts to escape the house. She is quickly killed after running into a trip wire strung outside the front door, cutting her throat.
Aubrey, upset by the death of her daughter, goes upstairs to her bedroom to grieve, but is murdered by an assailant under the bed. Hearing her scream, the family runs upstairs to find her corpse. On the wall, written in blood, are the words "You're Next". Staying in the room, Kelly (Margaret Laney) finds the killer still under the bed and runs to the neighbors' house, where she discovers the body of the man from the opening. She is then killed with an axe to the head.
Believing the killers have left, Crispian flees the house. Erin is attacked but kills her assailant with a meat tenderizer to the head. Paul discovers upstairs that the killers had been in the house before the family arrived, but he is murdered while trying to warn his kids. It is then revealed that his son Felix (Nicholas Tucci) and Felix's girlfriend, Zee (Wendy Glenn), are in on the murders, hoping to inherit the family's fortune.
Erin sets up small traps around the house hoping to catch the killers. She confides to Zee that she was raised by survivalists, a secret she kept hidden even from Crispian. Drake awakens and is almost killed, but Erin saves him, and the killer flees. Believing there should be weapons in the basement, Erin goes down with Drake, Zee, and Felix.
Erin and Zee leave the basement, leaving Felix and Drake alone. Felix kills Drake by impaling him with screwdrivers. Erin goes upstairs to try and find Paul and she is attacked by one of the killers, but manages to escape. She runs into the woods and is attacked again, but makes it back to the house. Once inside, she overhears a conversation between Felix, Zee, and two others, discovering their involvement. Her phone goes off, giving away her hiding spot, but also alerting her that her previous 911 text was received.
Erin kills the final two masked attackers, and sets up another trap with an axe above the front door. Erin is attacked by Felix and Zee, and she kills them after a long fight in the kitchen. Felix's phone rings and Erin answers without revealing her identity. Crispian is on the other line, revealing his involvement in the plot to kill the family. Crispian returns to the house and tries to explain his actions, but Erin kills him with a knife to the neck and eye.
A policeman arrives and upon seeing Erin stab Crispian, shoots her in the shoulder. He calls for backup and goes into the house. However, he enters through the booby-trapped front door despite Erin's attempts to warn him. As the axe hurtles towards his face, the film smash cuts to the end credits with the words "YOU'RE NEXT" written in blood.

Gary King (Simon Pegg), a hedonistic alcoholic, resolves to track down his estranged friends and complete the Golden Mile, an infamous pub crawl encompassing 12 pubs in their hometown of Newton Haven. The group attempted the crawl as teenagers over 20 years earlier, but failed to reach the final pub, The World's End.
Gary persuades Peter Page (Eddie Marsan), Oliver "O-Man" Chamberlain (Martin Freeman), Steven Prince (Paddy Considine) and Andie Knightley (Nick Frost) to join him in Newton Haven. While his friends have adult lives and responsibilities, Gary has changed little since 1990, remaining untrustworthy and impulsive. In the first pub, the First Post, the group and their mission are not met with the hero's welcome Gary expects. They are briefly joined at the Old Familiar by Oliver's sister Sam (Rosamund Pike), who rejects a sexual advance from Gary. Steven has always loved Sam, and has never forgiven Gary for sleeping with her many years ago. Gary is thrown out of the Famous Cock, having been permanently banned from it, but drinks some leftover beer on a table outside to satisfy his goal of drinking at every pub.
In the Cross Hands, Gary gets into a fight with a local teenager and is surprised at his strength and agility. Gary accidentally knocks his head off, exposing him as a robot. Gary's friends join him for another fight with more robot youths, after which Andy abandons his teetotaling ways and drinks an order of whiskey shots meant for all five. Unsure how many robots there may be, they decide to continue the pub crawl Gary had announced to the locals so as to avoid attracting suspicion.
At the Two Headed Dog, the group meets Sam and Gary warns her of the robot invasion. At the Mermaid, robots attempt to seduce the group and steal their DNA. When Sam's childhood crush Adrian appears, she panics and drags the others from the pub, explaining that Adrian had died in a motorcycle crash years previously. At the next pub, the Beehive, Guy Shepherd (Pierce Brosnan), a teacher at their school, encourages the group to accept their fate and become robots. The reappearance of a long-since-removed birthmark on Oliver's face makes the group realise he has been replaced with a robot based on his DNA, initiating a brawl. Despite the danger, Gary insists on drinking a pint during the fight. The group scatters, though Gary gets Sam to her car and tells her to leave Newton Haven.
Once reunited, the friends accuse each other of being replaced with robots like Oliver. Steven, Peter and Andy prove themselves with scars from their childhood, but Gary refuses to roll his sleeves up to reveal his elbow scar. Instead, he repeatedly headbutts a pillar to prove that his skull is tougher than the robots'. The robots close in on the group and capture Peter, but Gary is determined to finish the pub crawl and runs towards the final pub. Abandoning Steven, Andy chases Gary through the streets, fighting hordes of robots along the way.
At The World's End, Andy confronts Gary. He has never forgiven Gary for abandoning him when he saved Gary's life after a drug overdose, leaving Andy to be arrested. They begin to fight; Gary's arms are revealed to be bandaged after a recent suicide attempt. Gary tearfully tells Andy that finishing the Golden Mile is the only thing that matters in his life, and that he is jealous of Andy for having an ideal life. Andy reveals that his marriage has broken down.
Ignoring Andy's pleas, Gary tries to draw himself a final pint at the bar. The tap lever instead lowers the bar into a hidden chamber, where Steven, having escaped the robots, finds them. An alien (voiced by Bill Nighy) explains that the robot invasion of the town is one of several on Earth, as part of a "civilising" process for humanity's own good, including the great advance in telecommunications in recent decades. The aliens offer eternal youth as robots, but the three friends decline, arguing that humans should be allowed to be imperfect. Seeing that humans are more trouble than they are worth, the aliens agree to leave the planet and abandon the invasion. Sam arrives to drive them to safety as the town begins to self-destruct.
Some time later, Andy is recounting this story around a campfire in the ruins of London, explaining how the aliens' withdrawal from the planet was accompanied by a worldwide pulse that destroyed modern technology, setting humanity back to the Dark Ages. The abandoned robots have reactivated and are regarded with mistrust by surviving humans. Andy's marriage has recovered, Steven is in a relationship with Sam, and Peter and Oliver, though now robots, have returned to a semblance of their old lives. In the ruins of Newton Haven, the now-sober Gary enters a pub with robot duplicates of his younger friends and orders five glasses of water. When the bartender refuses to serve any non-humans, Gary draws a sword and leads his robot friends into a brawl.
